I clicked on the button to accept the next quest, but instead of giving me a new item to get, i got a message saying

I am not ready to give you another quest yet!
Come back later and I will see what I can do...

I haven't done her quest today. Do I need to wait 24hours from the last time I did one?

Even if you click a wrong button (the 'I don't have it' button) you should still have the quest there if you return before the time runs out. I hope you don't have to start over

No item, no timer, just come back later.

I submitted a bug report, but I'd hate to lose all those 25 levels

You have to wait 12 hours to get a new quest, not the next day. Hopefully this solves your problem...
