Yes. It's a one time use item.
How else could it work. Magical Plushies would be way too valuable otherwise

When you use them, they turn into the non magical version of the plushie

So, as an example (true story too) I had a grundo of some sort and I had a really bad computer. I played homby and the Fungus Balls really slowly (and this is when they gave away the plusies) and I won a Magical Chomby Plushie.
I used the "Magical Blue Chomby Plusie" on my "Purple Grundo" and it turned into a "Blue Chomby". My "Magical Blue Chomby Plushie" then turned into a "Blue Chomby Plushie.
I hope that made sense.
